Focal adhesions are identified as areas within the plasma membrane of tissue culture cells that adhere tightly to the underlying substrate. In vivo, these regions are involved in the adhesion of cells to the extracellular matrix. Paxillin and vinculin are cytoskeletal, focal adhesion proteins that are components of a protein complex which links the Actin network to the plasma membrane. Vinculin binding sites have been identified on other cytoskeletal proteins, including talin and alpha-actinin each contain actin binding sites. Reportedly, vinculin is a potential plasma biomarker for Age-related Macular Degeneration (AMD). The early detection of AMD using novel plasma biomarkers with genetic modeling may enable timely treatment and vision preservation in the elderly. Biotium Cytokeratin 10 (Suprabasal Epithelial Marker) (DE-K10), CF488A conjugate, 0.1mg/mL
This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Expression of CK10 is related to maturation of malignant keratinocytes, being preferentially detected in more-differentiated parts.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. IgM antibody is prominent in early immune responses to most antigens. Aberrant levels are associated with immune deficiency states, hereditary deficiencies, myeloma, Waldenstrom's macroglobulinemia, chronic infection and hepatocellular disease. This MAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The LMO2 protein has a central and crucial role in hematopoietic development and is highly conserved. It has a particular function in normal and lymphatic endothelial cells involving the regulation of angiogenesis and lymph-angiogenesis. Immunohistochemical studies have also demonstrated expression of LMO2 in both normal germinal center B-cells and germinal center-derived B-cell lymphomas, including follicular lymphoma and diffuse large B-cell lymphoma. The use of anti-LMO2 is valuable as a tool in the identification of lymphomas of B-cell origin. LMO2 is useful in differentiating follicular lymphoma (LMO2 ) from nodal marginal zone lymphoma (LM02-). It also is positive in Hodgkin's and Burkitt's lymphomas. B-cell lymphoma/leukemia 10 (BCL10) is a signaling protein that promotes apoptosis, pro-caspase-9 maturation, and activation of NF-kappa-B via NIK and IKK. BCL10 contains an N-terminal caspase recruitment domain (CARD), which is found in a number of apoptotic regulatory molecules. BCL10 is involved in the adaptive immune response, and may be an adapter protein between upstream TNFR1-TRADD-RIP complex and the downstream NIK-IKK-IKAP complex. This antibody recognizes an intermediate filament protein (IFP) of 55 kDa, which is identified as cytokeratin 7. This MAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract.
